    We evaluate the vascular irritation and active systemic allergy of the radiodiagnostic drug Technetium 99mTc-GSA injection,to provide reference for the clinical safety application of radiopharmaceutical drugs. The results show that there is no vascular irritation reaction after intravenous injection,and strong positive allergic reactions can be avoided by single clinical use.

    Based on the LNCaP cell-specific membrane antigen target for human prostate cancer,an equal amount of 125I-labelled reference compound 125I-A was added to the porous filter plate,and then different concentrations of ligands to be tested were added. With competitive binding assays,the affinity between drugs and targets wasstudied. We thus establish a screening method for targeted drugs based on isotope labelling invitro.

    Using 16srDNA technique,the dose effect of whole body exposure to 60Coγ-rays on the intestinal tissue and faecal flora of Beagles was analysed. The results showed that different doses of 60Co could change the structure of fecal microflora,and promote or aggravate intestinal damage,of Beagles.

    After systemic unilateral exposure to 60Co with different doses,the changes of platelets and coagulation function in beagles were observed at different times. The results showed that both low and high dose could activate endogenous coagulation mechanism,thrombosis and fibrinolysis hyperactivity,and the mechanism of bleeding death induced by high dose was related to the disorder of blood coagulation,anticoagulation and fibrinolysis system.

    With high-dose X-rays given twice to SD rats locally in singlepass,we explore to establish a radio-proctitis model in rats,via observing the clinical symptoms and histopathological changes. It shows that acute radio-proctitis in rats can be induced successfully by a single local X-ray exposure of 18 Gy. Our model is suitable for the study of the therapeutic effect of radiorectal damage drugs.

    The protective effect of oligochitosan on radiation damage in mice exposed to gamma rays was explored by means of flow cytometry,RT-PCR,Western blotting. The changes in bone marrow nucleated cells,lymphocyte apoptosis,spleen organ coefficient,cell apoptosis-related genes,P53 protein and GADD45 protein were tested. As compared with the control group,γ-ray exposed mice with a gavage of oligochitosan showed an improved survival rate,increase numbers of bone marrow nucleated cells,reduced numbers of apoptotic cells in bone marrow lymphocytes. For the test group,oligochitosan was observed to be able to down-regulate Bax,caspas-3 and up-regulate Bc1-2 for apoptosis-related genes,which give rise to anincreased Bcl-2/Bax ratio,and inhibite expression of P53 protein and GADD45 protein also. It suggests that oligochitosan may inhibit the expression of P53 protein and GADD45 protein,affect the expression of apoptosis-related genes,inhibit the production of apoptotic cells,and promote haematopoietic function recovery,and thus protect mice from radiation damage.

    The concentration of capecitabine and 5-fluorouracil in blood and the concentration of capecitabine in the intestinal single-pass perfusion effluent were tested by UPLC-MS/MS after single exposure of SD rats by linear accelerator.Cmax,Tmax,t1/2,AUC,absorption rate constant (Ka) and apparent absorption coefficient(Papp) were then calculated. It showes that the testing method is simple and reproducible,and can simultaneously determine the prototype drug capecitabine and active metabolite 5-fluorouracil in rat plasma. Our analysis indicate that the single exposure of 10 Gy in the abdomen of linear accelerator has inhibited the absorption of capecitabine into the blood circulation in the small intestine and colon,resulting in delayed peak reaching time,decreased peak concentration and blood drug concentration,and accelerated excretion,while capecitabine metabolic rate hasn’tbeen affected.

    Arrowsmith tool was used to explore the potential association between hypothermia therapy and radiation damage by taking PubMed literature as the analysis object. The results showed that there are about 30 keywords with extremely high correlation,and the top 5 are akt,pi3k,trpv1,toll receptor,Caspase-3 gene targets.
