12C离子辐射对人淋巴细胞端粒长度的影响

12C Irradiation Inducing Telomere Extension in HumanLymphocytes

摘要: 目的 探讨12C离子辐射对人淋巴细胞端粒长度的影响。方法 培养人淋巴细胞Peng-EBV接受12C离子束照射,剂量率0.3~0.5 Gy/min,照射剂量分别为0、0.1、0.5、1.0、2.0 Gy。在照射后24、48、72小时收集细胞,提取基因组DNA,采用荧光定量PCR方法检测样本相对端粒长度。结果 与对照组相比,照射后48和72小时,各剂量组受照细胞相对端粒长度增加显著。结论 12C离子照射可导致培养人淋巴细胞端粒延长,其中1.0 Gy组,照射后24、48、72小时都显著增加,且增加幅度大于0.1、0.5和2.0 Gy组的受照细胞。

Abstract: This paper is to investigate the effect of12C ion irradiation on telomere length in culture human lymphocytes. Culture human lymphocyte line Peng-EBV were exposed to 12C at 0.3-0.5 Gy/min at doses of 0,0.1,0.5,1.0 and 2.0 Gy, respectively. The cells were collected at 24, 48, 72 hours after exposure, respectively. Genome DNA was extracted. Relative telomere length was measured by fluorescence quantitative PCR. The relative telomere length obviously increased at 48 and 72 hours in each dose group for exposed cells. 12C ion irradiation may all result in telomere extension in culture human lymphocytes.
